SP1: Transcriptional analysis in defined cell types and single cells

This subproject provides bioinformatics and data management of sequencing data produced by all members of this CRU using a Bioinformatics Workflow System (BWS). The BWS will be updated to support still-maturing bioinformatics methods, such as single cell RNA-seq experiments and yet unsupported pipelines for the analysis of CRISPR library screens. The BWS serves as a data repository, enabling reproducible and secure sharing of bioinformatics analysis among all members of the CRU. Additionally, this project will create and curate a database with genome wide transcriptional profiles of Myeloproliferative Neoplasms (MPN-DB)in cells from mouse models, induced pluripotent stem cells and MPN patients with distinct mutations (JAK2, CALR, MPL) and from distinct MPN entities. The MPN-DB will be enhanced with single cell RNA sequencing from selected mouse models and patient samples. This will allow the unique characterization of rare stromal cells and the influence of MPN mutations in fibrosis-associated stromal cells.
